A study published in the journal Science Translational Medicine demonstrates that artificially recreating a rare genetic condition can give humans near-universal protection against viruses.

People with ISG15 protein deficiency experience a persistent mild inflammatory response, which, contrary to expectations, brings not only risks, but also a unique benefit - the ability to effectively fight all known viruses, including influenza and SARS-CoV-2. Scientists have found that this effect is due to the activation of 10 specific proteins that block viral replication.

Based on these data, an experimental mRNA-based therapy has been developed. It is a complex of 10 mRNAs enclosed in lipid nanoparticles that are delivered to the lungs through a nasal drip method. In tests on mice and hamsters, the therapy showed high effectiveness: it prevented viral replication and reduced the severity of the disease. Moreover, in cell culture, scientists could not find a single virus that could overcome the protection of this technology.

A particular advantage of the therapy is that it does not cause long-term inflammation, as in patients with natural ISG15 deficiency, but only briefly activates the immune system. At the same time, a person retains the ability to form his own immunity to specific pathogens.
